Abstract NoteIn recent years, the fabrication of nanomaterials and exploration of their properties have attracted the attention of various scientific disciplines such as biology, physics, chemistry, and engineering. Although nanoparticulate systems are of significant interest in various scientific and technological areas, there is little known about the safety of these nanoscale objects. It has now been established that the surfaces of nanoparticles are immediately covered by biomolecules (e.g. proteins, ions, and enzymes) upon their entrance into a biological medium. This interaction with the biological medium modulates the surface of the nanoparticles, conferring a ???biological identity??? to their surfaces (referred to as a ???corona???), which determines the subsequent cellular/tissue responses. The new interface between the nanoparticles and the biological medium/proteins, called ???bio-nano interface,??? has been very rarely studied in detail to date, though the interest in this topic is rapidly growing. In this book, the importance of the physiochemical characteristics of nanoparticles for the properties of the protein corona is discussed in detail, followed by comprehensive descriptions of the methods for assessing the protein-nanoparticle interactions. The advantages and limitations of available corona evaluation methods (e.g. spectroscopy methods, mass spectrometry, nuclear magnetic resonance, electron microscopy, X-ray crystallography, and differential centrifugal sedimentation) are examined in detail, followed by a discussion of the possibilities for enhancing the current methods and a call for new techniques. Moreover, the advantages and disadvantages of protein-nanoparticle interaction phenomena are explored and discussed, with a focus on the biological impacts

Abstract NoteThis book addresses the possibilities and challenges in mimicking biological membranes and creating membrane-based sensor and separation devices. It covers recent advances in developing biomimetic membranes for technological applications with a focus on the use of integral membrane protein mediated transport. It describes the fundamentals of biosensing as well as separation and shows how the two processes??work together in biological systems. The book provides an overview of the current state of the art, points to areas that need further investigation and anticipates future directions in the field. Biomimetics is a truly cross-disciplinary approach and this is exemplified by the challenges in mimicking osmotic processes as they occur in nature using aquaporin protein water channels as central building blocks. In the development of a biomimetic sensor/separation technology, both channel and carrier proteins are important and examples of how these may be reconstituted and controlled in biomimetic membranes are presented. Also new developments in our understanding of the reciprocal coupling between the material properties of the biomimetic matrix and the embedded proteins are discussed.?? The basic concepts of membrane barrier properties are introduced and discussed in terms of lipid and polymer based membranes. Once a given protein is reconstituted in its final host biomimetic matrix, its stability needs to be maintained and controlled and the challenges associated with insertion and stabilization of alpha-helical bundle proteins are exemplified with aquaporin and ion channels as well as sodium-potassium ATPase proteins. The concept of multi-scale modeling is introduced and exemplified by the use of molecular dynamics, dissipative particle dynamics, and computational fluid dynamics simulations illustrating the issues involved in developing and describing biomimetic systems in a wide range of time and length scales. Scalability is a general issue for all nano-inspired technology developments and many biomimetic membrane applications require that the device can be used in the macroscopic realm. This challenge is addressed here in the context of fabricating biomimetic components, membrane arrays, and compartmentalized systems together with the challenges related to microfluidic design strategies for biomimetic device developments

Abstract NoteSpectroscopic methods have opened up a new horizon in our knowledge of solid-state materials. Numerous techniques using electromagnetic radiation or charged and neutral particles have been invented and worked out to a high level in order to provide more detailed information on the solids. The text presented here is an updated description of such methods as they were originally presented in the first edition. It covers linear response of solids to electromagnetic radiation in a frequency range extending from megahertz or gigahertz as used in spin resonance spectroscopy, to infrared spectroscopy and various forms of spectroscopy in the visible and near visible spectral range. It extends to spectroscopy in the UV and x-ray spectral range and eventually several spectroscopic methods are addressed in the frequency range of g radiation. Likewise linear response to irradiation with particles such as electrons, positrons, muons, neutrons, and atoms is discussed. Instrumental and technical background is provided as well as application to the analysis of the solid state. New developments especially addressed in this second edition cover the exciting new field of mesoscopic and nanoscopic solids using light scattering from small particles, single electron (addition) spectroscopy, and quantization in magnetic fields. Likewise, the development and application of synchrotron radiation has encountered a dramatic progress particularly in the field of high resolution and angular resolved photoelectron spectroscopy. Free electron lasers from undulators in linear accelerators are becoming available with dramatic enhancement in brightness and provide a fourth generation of synchrotron light sources. The better understanding and the possibilities to manipulate spin states as it happens in spintronic research stimulated sophisticated technologies in electron and nuclear spin resonance which are also covered in the new edition. Breakthrough developments were obtained in the field of light emission from semiconductor devices and for femtosecond spectroscopy. In addition in almost all other spectroscopic techniques progress was obtained in the last years and is reviewed in the text. Thus the book may be considered as a state-of-the-art introductory text in solid-state spectroscopy. Each chapter includes problems. Abstract NoteIn today???s world, three great classes of non-infectious diseases ??? the metabolic syndromes (such as type 2 diabetes and atherosclerosis), the cancers, and the neurodegenerative disorders ??? have risen to the fore. These diseases, all associated with increasing age of an individual, have proven to be remarkably complex and difficult to treat. This is because, in large measure, when the cellular signaling pathways responsible for maintaining homeostasis and health of the body become dysregulated, they generate equally stable disease states. As a result the body may respond positively to a drug, but only for a while and then revert back to the disease state. Cellular Signaling in Health and Disease summarizes our current understanding of these regulatory networks in the healthy and diseased states, showing which molecular components might be prime targets for drug interventions. This is accomplished by presenting models that explain in mechanistic, molecular detail how a particular part of the cellular signaling web operates properly in health and improperly in disease. The stability of the health- and disease-associated states is dynamic and supported by multiple feedback loops acting positively and negatively along with linkages between pathways. During the past few years an ongoing series of important discoveries have been made that advance our understanding of how the body works and may guide us on how to better deal with these diseases. These include the discovery of chronic inflammation as a causal factor in all of these disease classes, the appearance of reactive oxygen species as a messenger molecule that can act both positively and negatively, the propensity of proteins to misfold into aggregation- and disease-prone forms, and the rise of epigenetics including the emergence of small non-coding RNA with important regulatory functions out of the so-called junk RNA.
